Injection site vaccinology of a recombinant vaccinia-based vector reveals diverse innate immune signatures

چکیده

Poxvirus systems have been extensively used as vaccine vectors. Herein a RNA-Seq analysis of intramuscular injection sites provided detailed insights into host innate immune responses, well expression vector and recombinant immunogen genes, after vaccination with new multiplication defective, vaccinia-based vector, Sementis Copenhagen Vector. Chikungunya Zika virus mRNA protein was associated necrosing skeletal muscle cells surrounded by mixed cellular infiltrates. The multiple adjuvant signatures at 12 hours post-vaccination were dominated TLR3, 4 9, STING, MAVS, PKR the inflammasome. Th1 cytokine IFN?, TNF IL1?, chemokine CCL5 CXCL12. Multiple dendritic cell stimulation evident. By day seven, transcripts absent, death, neutrophil, macrophage inflammation annotations had abated. No compelling arthritis identified. Such site vaccinology approaches should inform refinements in poxvirus-based design.

Systems vaccinology: probing humanity's diverse immune systems with vaccines.

Homo sapiens are genetically diverse, but dramatic demographic and socioeconomic changes during the past century have created further diversification with respect to age, nutritional status, and the incidence of associated chronic inflammatory disorders and chronic infections. These shifting demographics pose new challenges for vaccination, as emerging evidence suggests that age, the metabolic ...

Construction of a recombinant vector for site-directed mutagenesis in Salmonella typhimurium

BACKGROUND: Among all common techniques in sitedirectedmutagenesis, λ Red recombinase system has beenwidely used to knock out chromosomal genes in bacteria. In thismethod, there is always the risk of DNA Linear digestion byhost's restriction enzymes that leads to the low frequency ofrecombination. OBJECTIVES:To overcome this, we constructeda recombinant vector to disrupt phoP gene in Salmonella...

Successive site translocating inoculation potentiates DNA/recombinant vaccinia vaccination

DNA vaccines have advantages over traditional vaccine modalities; however the relatively low immunogenicity restrains its translation into clinical use. Further optimizations are needed to get the immunogenicity of DNA vaccine closer to the level required for human use. Here we show that intramuscularly inoculating into a different limb each time significantly improves the immunogenicities of b...

Injection site lipoatrophy: a rare complication of recombinant human insulin.

Fig. 1 and Fig. 2 : Lipoatrophy over the left para-umbilical region. A 59-year-old male with type 2 diabetes of 19 years duration had been treated with premixed recombinant human insulin (30% regular and 70% NPH) for the last 18 months. He was injecting himself predominantly over the left side of the abdomen. He developed subcutaneous lipoatrophy around the injection site about 14 months after ...
